This method works because PEG can make cell membranes soft and let them merge. Data shows PEG1500 at 30-50% concentration can help 80% cells fuse. Centrifuge speed 1000-1500rpm for 15 minutes separates hybrids better. Enzymes like trypsin remove non-fused cells. So step by step, first fuse then separate. But if centrifuge time is too short, small cells might mix again. Also if PEG is too much, it could kill cells. So balance is key. Like mix 1ml PEG with 9ml cells, wait 5 minutes, then spin. This way most cells become hybrids but not damaged.
